Design Notes

CusR is a response regulator that binds to DNA when phosphorylated. This small part is the recognition squence for the protein and we designed it to be inserted downstream of a promoter to allow negative regulation of transcription by CusR phosphorylation. In common with many response regulators we expect over-expression of CusR to result in binding and so negative regulation of an upstream promoter. These negative effects are expected through bound CusR physically blocking RNA polymerase. This part is complementary to part BBa_K1349003.

A construct with this part between a promoter and a gene is expected to show negative regulation of the gene by phosphorylation of CusR (or overexpression of CusR).
